This Notice of Funding Opportunity (NOFO) invites applications proposing a Data Coordinating Center that integrates and disseminates multiscale mouse brain datasets for the aging and Alzheimer's disease (AD) research community. The Data Coordinating Center will establish the Aging and AD Mouse Brain Atlas (AD-MBA), which will serve as a central, unified research resource for multi-omic and connectomic mouse brain datasets emerging from the NIA AD Multimodal Atlas Projects (AD-MAPs). The Data Coordinating Center will (1) leverage the NIA-supported AD Knowledge Portal data infrastructure to facilitate hosting, deposition, and dissemination of multimodal datasets generated by the AD-MAPs program, (2) facilitate collaborative analyses among the data-contributing teams, (3) develop/optimize and incorporate tools for multiscale integration of multi-omics and connectomics datasets, and (4) create a broadly accessible web-based interface for the visualization and dissemination of integrated multiscale data.

Section I. Notice of Funding Opportunity Description

Background

Alzheimer's disease (AD) is a progressive neurodegenerative disease of the brain and the most common form of dementia in older adults. It is estimated that AD starts 15–20 years before the appearance of the onset of clinical symptoms and ultimately leads to a loss of memory and cognitive skills and a decline in quality of life in older adults. The progression of AD often follows a predictable temporal and spatial pattern where specific brain regions are affected at different stages of the disease. However, it is unclear why the disease affects certain regions of the brain or which specific cell types within these brain regions are selectively vulnerable.

To better understand the mechanisms and impact of selectively vulnerable brain regions, cell types, and circuits in AD progression, NIA established the AD Multimodal Atlas Projects (AD-MAPs). The goal of AD-MAPs is to generate comprehensive multimodal atlases and brain-wide connectomes across species as a platform to discover cell types and circuits that are altered in normal aging and AD progression. In addition, the AD-MAPs program supports projects performing secondary analyses of these datasets.

One focus of the AD-MAPs program is to characterize cells and circuits in mouse models of aging and AD. These mouse model studies are performing high-resolution transcriptomic and epigenomic profiling, as well as executing spatial transcriptomics and circuit-level mapping. These projects also incorporate neuropathological data across multiple mouse models of aging and AD. Many of these datasets have the potential to be integrated and shared as a common resource, increasing the overall value of each individual project by enhancing adherence to FAIR (Findable, Accessible, Interoperable, Reuseable) data principles. However, these large projects remain siloed, and there is a need for a centralized data sharing infrastructure to integrate and disseminate these valuable datasets.

Research Objectives

This NOFO invites applications proposing a Data Coordinating Center that will develop a centralized resource for discovery, access, and integration of mouse model datasets generated by projects in the AD-MAPs program. A successful Data Coordinating Center will implement open science principles and FAIR data practices to integrate disparate multiscale datasets from across projects and create the Aging and AD Mouse Brain Atlas (AD-MBA) as a robust resource for the research community.

The Data Coordinating Center will take advantage of the unique opportunity to unify aging and AD mouse brain datasets spanning multi-omics and connectomics. This multiscale integration builds on a key takeaway from an NIA-hosted workshop in March 2023 that emphasized the value of incorporating data across scales to better understand aging and AD/ADRD and identified this gap as "the next big opportunity" facing the field. Furthermore, current advances in technology have enabled multiscale integration of mouse brain data spanning single-cell transcriptomics to circuit-level connectomics.

This NOFO is intended to support the creation of a centralized infrastructure that brings together multi-omics and connectomics mouse data for the AD-MBA, a first-of-its-kind data resource for the aging and AD research community. To create the AD-MBA, a Data Coordinating Center is needed to:

  1. Leverage the NIA-supported AD Knowledge Portal FAIR data infrastructure to facilitate hosting, deposition, and dissemination of multimodal datasets generated by the AD-MAPs program.
  2. Facilitate collaborative analyses among the data-contributing teams.
  3. Develop/optimize and incorporate open-source tools for multiscale integration of multi-omics and connectomics datasets.
  4. Create open-source, web-based interfaces for visualization query of integrated multiscale data.

The Data Coordinating Center will achieve two major goals:

1. Unify disparate, multimodal data generated in brain tissue of mouse models of aging and AD by aggregating and harmonizing datasets across at least 5 projects in the AD-MAPs program.

  • The Data Coordinating Center must aggregate and harmonize multiscale data from multiple mouse models of aging and AD, including different AD pathologies, multiple brain regions, both sexes, and early to late-stage AD progression. These separate projects are collecting datatypes spanning genomics and connectomics.
  • The Data Coordinating Center must develop/optimize and share tools for multiscale integration of multi-omics and connectomics datasets.

2. Create a community portal that will serve as a knowledge base for discovery and enable access to the datasets from the contributing projects from the AD-MAPs program, the associated open-source tools, and web-based interfaces for data visualization and query.

  • The Data Coordinating Center must facilitate the hosting, deposition, and dissemination of multimodal datasets generated by the contributing AD-MAPs teams.
  • The data repository for the raw, processed, and harmonized datasets must be the NIA-supported AD Knowledge Portal. The AD Knowledge Portal is a FAIR data infrastructure containing both human and animal datasets from over a dozen NIA-supported systems biology consortia and translational centers such as AMP-AD and MODEL-AD.
  • The Data Coordinating Center applicants are strongly encouraged to complement and align with existing NIA/NIH data resources, such as those produced by the MODEL-AD consortium and those produced by the BRAIN Initiative.

Specific Areas of Research Interest

Activities appropriate for the AD-MBA Data Coordinating Center include, but are not limited to the following examples:

  • Develop the AD-MBA as a public portal for the broad sharing of raw and processed data, multiscale models, and analytical tools.
  • Coordinate timely deposition and release of newly generated datasets contributed by AD-MAPs members.
  • Coordinate and provide adequate annotation and curation of datasets to enable data integration and harmonization and to maximize data usability.
  • Facilitate collaborative analyses among AD-MAPs data-contributing teams that will leverage combined resources across partners to more effectively answer pressing scientific questions.
  • Develop web-based, open-source data visualization interfaces and tools to enable data scientists and biologists to query and interact with the rich datasets and multiscale models being developed by the AD-MAPs program.

Applicants are strongly encouraged to consult with the NIA Scientific Contact early in the pre-submission process to ensure that the application is responsive to the programmatic goals of this NOFO.

Applications Not Responsive to this NOFO

The following types of applications are not responsive to this NOFO and will not be reviewed:

  • Applications that do not propose to aggregate and harmonize datasets across at least five projects in the AD-MAPs program.
  • Applications that do not propose to integrate data types including, at minimum, single-cell/nucleus transcriptomics and connectomics.
  • Applications that do not propose to create an accessible community portal to enable broad dissemination.
  • Applications that do not propose to develop, optimize, and/or utilize appropriate computational tools to integrate datasets spanning modalities (genomics and connectomics).
  • Applications that propose to incorporate datasets not collected by projects in the AD-MAPs program.
  • Applications that propose to directly acquire new primary data.
  • Applications that propose to integrate data that were not generated from mouse models.

Funding Instrument

Cooperative Agreement: A financial assistance mechanism used when there will be substantial Federal scientific or programmatic involvement. The renewal process may require as much time as the initial registration. SAM registration includes the assignment of a Commercial and Government Entity (CAGE) Code for domestic organizations which have not already been assigned a CAGE Code. Foreign organizations must obtain a NATO Commercial and Government Entity (NCAGE) Code (in lieu of a CAGE code) in order to register in SAM.
    • Unique Entity Identifier (UEI)- A UEI is issued as part of the SAM.gov registration process. The same UEI must be used for all registrations, as well as on the grant application.

Applicant organizations may submit more than one application, provided that each application is scientifically distinct.

Research Strategy

Applications must describe plans to achieve the following five objectives.

1) Aggregate and harmonize mouse brain multi-modal (genomics and connectomics) datasets emerging from at least 5 projects in the AD-MAPs program.

  • Applications must describe plans to coordinate with project PIs to ingest existing and future datasets and associated metadata from at least 5 projects in the AD-MAPs program.
  • Data types to be aggregated must include, at minimum, single-cell/nucleus transcriptomics and connectomics.
  • Plans must include harmonization of datasets within the same modality.
  • Applications must include plans to aggregate and harmonize data from multiple mouse models of aging and AD, including different AD pathologies, multiple brain regions, both sexes, and early to late-stage AD progression. Data types should include, but are not limited to, the following:

2) Develop and incorporate tools for multiscale modeling of multi-omics and connectomics datasets.

  • To achieve multiscale modeling, applications must include plans to develop, optimize, and/or utilize appropriate multiscale computational tools to integrate datasets spanning modalities (genomics and connectomics).
  • Datasets for multiscale integration must include, at minimum, single-cell/nucleus transcriptome and connectome. Inclusion of other modalities is strongly encouraged, such as epigenome, spatial transcriptome, and pathology.

3) Create an accessible platform to disseminate the data and tools to the research community: the AD-MBA.

  • Applications must propose a web-based interface that meets the needs of the research community and must serve as a knowledge base that includes the following:
    • Open-access tools to visualize and query harmonized, multiscale datasets, including methods developed for multiscale data integration in (2) above.
    • Access points for raw and processed datasets from the contributing projects.
  • Applications must describe plans to facilitate hosting, deposition, and dissemination of multimodal datasets generated by the contributing AD-MAPs teams.

4) Capitalize on existing resources and infrastructure.

  • The data repository for the raw, processed, and harmonized datasets must be the NIA-supported AD Knowledge Portal.
  • The web-based portal should be modeled after existing resources, including but not limited to NIA's MODEL-AD and/or the Allen Institute for Brain Science portals for the Allen Brain Cell Atlas and the Mouse Brain Connectivity Atlas for normal mouse brain.
  • Applications are strongly encouraged to complement and align with existing NIA/NIH data resources, such as those produced by the MODEL-AD consortium and those produced by the BRAIN Initiative.

5) Evaluation Plan

  • Applications must include a well-defined plan for evaluating the activities supported by the award. The evaluation plan must include specific and feasible measures of progress and a timeline with major milestones. The measures of progress included should assess the administrative and scientific accomplishments, both yearly and overall. The milestones must include clear outcomes and actionable criteria for evaluating progress and decisions on project continuation and direction. Plans for ongoing feedback and critical evaluation by NIH Program Staff should be included.

Letters of Support

Applications must provide letters of support from the PIs of each project proposed for integration into the AD-MBA.

Applicants are encouraged to submit applications before the due date to ensure they have time to make any application corrections that might be necessary for successful submission. When a submission date falls on a weekend or Federal holiday, the application deadline is automatically extended to the next business day.

Organizations must submit applications to Grants.gov (the online portal to find and apply for grants across all Federal agencies). Applicants must then complete the submission process by tracking the status of the application in the eRA Commons, NIH's electronic system for grants administration. Section V. Application Review Information

1. Criteria

Only the review criteria described below will be considered in the review process.  Applications submitted to the NIH in support of the NIH mission are evaluated for scientific and technical merit through the NIH peer review system.

Overall Impact

Reviewers will provide an overall impact score to reflect their assessment of the likelihood for the project to exert a sustained, powerful influence on the research field(s) involved, in consideration of the following review criteria and additional review criteria (as applicable for the project proposed).

Scored Review Criteria

Reviewers will consider each of the review criteria below in the determination of scientific merit and give a separate score for each. An application does not need to be strong in all categories to be judged likely to have major scientific impact. For example, a project that by its nature is not innovative may be essential to advance a field.

 

Does the proposed Center address the needs of the research projects that it will coordinate? Is the scope of activities proposed for the Center appropriate to meet those needs? Will successful completion of the aims bring unique advantages or capabilities to the research projects?

Specific to this NOFO:

  • How well does the proposed portal and knowledge base meet the needs of the research community?

 

Are the PD(s)/PI(s) and other personnel well suited to their roles in the Center? Do they have appropriate experience and training, and have they demonstrated experience and an ongoing record of accomplishments in managing collaborative research? Do the investigators demonstrate significant experience with coordinating collaborative basic research? If the Center is multi-PD/PI, do the investigators have complementary and integrated expertise and skills; are their leadership approach, governance, plans for conflict resolution, and organizational structure appropriate for the Center? Does the applicant have experience overseeing selection and management of subawards, if needed?

Specific to this NOFO:

  • How well do the letters of support from the PIs of the contributing AD-MAPs projects demonstrate feasibility of collaboration?

 

Does the application propose novel organizational concepts, management strategies, or instrumentation in coordinating the research projects the Center will serve? Are the concepts, strategies, or instrumentation novel to one type of research program or applicable in a broad sense? Is a refinement, improvement, or new application of organizational concepts, management strategies or instrumentation proposed?


 

Are the overall strategy, operational plan, and organizational structure well-reasoned and appropriate to accomplish the goals of the research resource the Center will serve? Will the investigators promote strategies to ensure a robust and unbiased scientific approach across the resource, as appropriate for the work proposed? Are potential problems, alternative strategies, and benchmarks for success presented? If the resource is in the early stages of operation, does the proposed strategy adequately establish feasibility and manage the risks associated with the activities of the resource? Are an appropriate plan for work-flow and a well-established timeline proposed? Have the investigators presented adequate plans to ensure consideration of relevant biological variables, such as sex, for studies of vertebrate animals or human subjects?

Specific to this NOFO:

  • How comprehensive are the plans to coordinate with multiple project PIs to ingest existing and future datasets and associated metadata?
  • How feasible are the plans to aggregate and harmonize data from multiple mouse models of aging and AD, including different AD pathologies, multiple brain regions, both sexes, and early to late-stage AD progression?
  • How well does the approach determine which datasets meet minimum standards for rigor and reproducibility to ensure that only data of sufficient quality is incorporated?
  • How feasible are the plans to harmonize datasets within the same modality?
  • How well does the approach develop, optimize, and/or utilize appropriate multiscale computational tools to integrate datasets spanning modalities (at minimum, single cell/nucleus transcriptome and connectome)?
  • How well formulated is the plan to disseminate the open-source, multiscale integration methodology?
  • How well does the proposed web-based interface create a knowledge base for visualizing and querying harmonized, multiscale datasets and enable access to both raw and processed datasets?
  • How well does the proposed platform facilitate hosting, deposition, and dissemination of multimodal datasets generated by the contributing AD-MAPs teams?
  • How specific and feasible are the measures of progress, timeline, and major milestones as defined in the Evaluation Plan?

 

Will the institutional environment in which the Center will operate contribute to the probability of success in facilitating the research projects it serves? Are the institutional support, equipment and other physical resources available to the investigators adequate for the Center proposed? Will the Center benefit from unique features of the institutional environment, infrastructure, or personnel? Are resources available within the scientific environment to support electronic information handling?


Additional Review Criteria

As applicable for the project proposed, reviewers will evaluate the following additional items while determining scientific and technical merit, and in providing an overall impact score, but will not give separate scores for these items.

 

For research that involves human subjects but does not involve one of the categories of research that are exempt under 45 CFR Part 46, the committee will evaluate the justification for involvement of human subjects and the proposed protections from research risk relating to their participation according to the following five review criteria: 1) risk to subjects, 2) adequacy of protection against risks, 3) potential benefits to the subjects and others, 4) importance of the knowledge to be gained, and 5) data and safety monitoring for clinical trials.

For research that involves human subjects and meets the criteria for one or more of the categories of research that are exempt under 45 CFR Part 46, the committee will evaluate: 1) the justification for the exemption, 2) human subjects involvement and characteristics, and 3) sources of materials. For additional information on review of the Human Subjects section, please refer to the Guidelines for the Review of Human Subjects.

 This NOFO only accepts applications that do not propose clinical trials. Note: Applications may propose activities involving human subjects that are not deemed clinical trials.


 

When the proposed project involves human subjects and/or NIH-defined clinical research, the committee will evaluate the proposed plans for inclusion. For additional information on review of the Inclusion section, please refer to the Guidelines for the Review of Inclusion in Clinical Research


 

The committee will evaluate the involvement of live vertebrate animals as part of the scientific assessment according to the following three points: (1) a complete description of all proposed procedures including the species, strains, ages, sex, and total numbers of animals to be used; (2) justifications that the species is appropriate for the proposed research and why the research goals cannot be accomplished using an alternative non-animal model; and (3) interventions including analgesia, anesthesia, sedation, palliative care, and humane endpoints that will be used to limit any unavoidable discomfort, distress, pain and injury in the conduct of scientifically valuable research. Methods of euthanasia and justification for selected methods, if NOT consistent with the American Veterinary Medical Association (AVMA) Guidelines for the Euthanasia of Animals, is also required but is found in a separate section of the application. For additional information on review of the Vertebrate Animals Section, please refer to the Worksheet for Review of the Vertebrate Animals Section.


 

Reviewers will assess whether materials or procedures proposed are potentially hazardous to research personnel and/or the environment, and if needed, determine whether adequate protection is proposed.


 

For Resubmissions (as applicable), the committee will evaluate the application as now presented, taking into consideration the responses to comments from the previous scientific review group and changes made to the project.


 

For Renewals (as applicable), the committee will consider the progress made in the last funding period.


 

For Revisions (as applicable), the committee will consider the appropriateness of the proposed expansion of the scope of the project. If the Revision application relates to a specific line of investigation presented in the original application that was not recommended for approval by the committee, then the committee will consider whether the responses to comments from the previous scientific review group are adequate and whether substantial changes are clearly evident.


Additional Review Considerations

As applicable for the project proposed, reviewers will consider each of the following items, but will not give scores for these items, and should not consider them in providing an overall impact score.

Reviewers will assess the information provided in this section of the application, including 1) the Select Agent(s) to be used in the proposed research, 2) the registration status of all entities where Select Agent(s) will be used, 3) the procedures that will be used to monitor possession use and transfer of Select Agent(s), and 4) plans for appropriate biosafety, biocontainment, and security of the Select Agent(s).


 

Reviewers will comment on whether the Resource Sharing Plan(s) (e.g., Sharing Model Organisms) or the rationale for not sharing the resources, is reasonable.


 

For projects involving key biological and/or chemical resources, reviewers will comment on the brief plans proposed for identifying and ensuring the validity of those resources.


 

Reviewers will consider whether the budget and the requested period of support are fully justified and reasonable in relation to the proposed research.


2. Review and Selection Process

Applications will be evaluated for scientific and technical merit by (an) appropriate Scientific Review Group(s) convened by the Center for Scientific Review (CSR), in accordance with NIH peer review policies and practices, using the stated review criteria. Assignment to a Scientific Review Group will be shown in the eRA Commons.

As part of the scientific peer review, all applications will receive a written critique.

Applications may undergo a selection process in which only those applications deemed to have the highest scientific and technical merit (generally the top half of applications under review) will be discussed and assigned an overall impact score.

Requests for reconsideration of initial peer review will not be accepted for applications submitted in response to this NOFO. 

Applications will be assigned to the appropriate NIH Institute or Center. Applications will compete for available funds with all other recommended applications submitted in response to this NOFO. Following initial peer review, recommended applications will receive a second level of review by the National Advisory Council on Aging. The following will be considered in making funding decisions:

  • Scientific and technical merit of the proposed project as determined by scientific peer review.
  • Availability of funds.
  • Relevance of the proposed project to program priorities.

If the application is under consideration for funding, NIH will request "just-in-time" information from the applicant as described in the NIH Grants Policy Statement Section 2.5.1. Just-in-Time Procedures. This request is not a Notice of Award nor should it be construed to be an indicator of possible funding.

Prior to making an award, NIH reviews an applicant's federal award history in SAM.gov to ensure sound business practices. An applicant can review and comment on any information in the Responsibility/Qualification records available in SAM.gov.  NIH will consider any comments by the applicant in the Responsibility/Qualification records in SAM.gov to ascertain the applicant's integrity, business ethics, and performance record of managing Federal awards per 2 CFR Part 200.206 "Federal awarding agency review of risk posed by applicants."  This provision will apply to all NIH grants and cooperative agreements except fellowships.

3. Anticipated Announcement and Award Dates

After the peer review of the application is completed, the PD/PI will be able to access his or her Summary Statement (written critique) via the eRA Commons. Refer to Part 1 for dates for peer review, advisory council review, and earliest start date.

Section VI. Award Administration Information

1. Award Notices

A Notice of Award (NoA) is the official authorizing document notifying the applicant that an award has been made and that funds may be requested from the designated HHS payment system or office. The NoA is signed by the Grants Management Officer and emailed to the recipient's business official.

In accepting the award, the recipient agrees that any activities under the award are subject to all provisions currently in effect or implemented during the period of the award, other Department regulations and policies in effect at the time of the award, and applicable statutory provisions.

Recipients must comply with any funding restrictions described in Section IV.6. Funding Restrictions. Any pre-award costs incurred before receipt of the NoA are at the applicant's own risk.  For more information on the Notice of Award, please refer to the NIH Grants Policy Statement Section 5. The Notice of Award and NIH Grants & Funding website, see Award Process.

Institutional Review Board or Independent Ethics Committee Approval: Recipient institutions must ensure that protocols are reviewed by their IRB or IEC. To help ensure the safety of participants enrolled in NIH-funded studies, the recipient must provide NIH copies of documents related to all major changes in the status of ongoing protocols.

2. Administrative and National Policy Requirements

The following Federal wide and HHS-specific policy requirements apply to awards funded through NIH:

All federal statutes and regulations relevant to federal financial assistance, including those highlighted in NIH Grants Policy Statement Section 4 Public Policy Requirements, Objectives and Other Appropriation Mandates.

By applying for or accepting federal funds from HHS, recipients certify compliance with all federal antidiscrimination laws and these requirements and that complying with those laws is a material condition of receiving federal funding streams. Recipients are responsible for ensuring subrecipients, contractors, and partners also comply.

Applicants and recipients are strongly encouraged to refer to the NIH Director's Statement of Priorities, entitled "Advancing NIH's Mission Through a Unified Strategy." 

Recipients are responsible for ensuring that their activities comply with all applicable federal regulations. Pursuant to 2 CFR 200.340, by accepting an NIH award, the recipient agrees that continued funding for the award is contingent upon the availability of appropriated funds, recipient satisfactory performance, compliance with the Terms and Conditions of the award, and may also otherwise be terminated, to the extent authorized by law, if the agency determines that the award no longer effectuates the program goals or agency priorities, in line with 2 CFR 200.340(a)(4).

Pursuant to the Cybersecurity Act of 2015, Div. N, § 405, Pub. Law 114-113, 6 USC § 1533(d), the HHS Secretary has established a common set of voluntary, consensus-based, and industry-led guidelines, best practices, methodologies, procedures, and processes.

Successful recipients under this NOFO agree that:

When recipients, subrecipients, or third-party entities have:

  • ongoing and consistent access to HHS owned or operated information or operational technology systems; and
  • receive, maintain, transmit, store, access, exchange, process, or utilize personal identifiable information (PII) or personal health information (PHI) obtained from the awarding HHS agency for the purposes of executing the award.

Cybersecurity plans and procedures must at minimum include the following:

  • Develop cybersecurity plans and procedures, modeled after the NIST Cybersecurity framework, to protect HHS systems and data:
    • Identify:
      • Develop an inventory of all assets and accounts with access to HHS owned and operated information or operational technology systems or which obtain PII or PHI for the purposes of the award.
    • Protect:
      • Limit access to HHS owned and operated systems to only those in need of access to complete reward activities.
      • Require all staff to complete annual cybersecurity and privacy awareness training. Visit 405(d): Knowledge on Demand (hhs.gov) to obtain free trainings, if needed.
      • Enable multifactor authentication for all employees, subrecipients, and third-party entities to access HHS owned and operated information or operational technology systems.
      • Regularly backup sensitive data and test backups.
    • Detect:
      • Install anti-virus or anti-malware software on all devices, servers, and accounts used to connect to HHS owned and operated systems.
    • Respond:
      • Develop an incident response plan. See Incident-Response-Plan-Basics_508c.pdf (cisa.gov) to learn about developing incident response plans.
      • Have cybersecurity incident reporting procedures that ensure the relevant HHS awarding agencies are notified of a cybersecurity incident within 48 hours of discovery. A cybersecurity incident is defined as an unplanned interruption to a technology service or reduction in the quality of a technology service, or an occurrence that actually or potentially jeopardizes the confidentiality, integrity, or availability of an information system or the information the system processes, stores, or transmits.
    • Recover:
      • Investigate incidents and plug any security gaps identified. 

All activities proposed in your application and budget narrative must align with applicable law, including but not limited to statutes, executive orders, federal regulations and applicable judicial holdings.  Accordingly, discretionary awards shall not be used to fund, promote, encourage, subsidize, or facilitate; racial preferences or other forms of racial discrimination by the recipient, including activities where race or intentional proxies for race will be used as a selection criterion for employment or program participation; denial by the recipient of the sex binary in humans, or the belief that sex is a chosen or mutable characteristic; illegal immigration; or any other initiatives that compromise public safety.  If an application does not align, the application will not receive funding to the extent permitted by law and applicable court orders.

For applications involving substance abuse, the application must not support harm reduction. Please see Updated Funding Guidance for Recipients on Supplies and Services.

For applications involving funding Medication-Assisted Treatment (MAT) or medications for opioid use disorder (MOUD), this funding should be used to provide comprehensive treatment and recovery support services rather than medication-only models for opioid use disorder. Services should include medications, where clinically indicated, in conjunction with psychosocial and other treatment and recovery support services. Funding can also be used to support individualized tapering and discontinuation of medications when clinically indicated. Please see Updated Funding Guidance for Recipients on  MAT/MOUD.

As of October 1, 2025, HHS has adopted 2 CFR Part 200, with some modifications included in 2 CFR Part 300. These regulations replace those in 45 CFR Part 75. However, for NIH, under the Consolidated Appropriations Act for FY 2026, (P.L. 119-75, Division B, Title II, Sec. 224), the provisions relating to indirect costs in 45 CFR 75 continue to apply to NIH awards. Consistent with the statute, NIH will not apply updated thresholds outlined within 2 CFR Part 200, at this time.

In administering programs under this and all funding announcements, NIH prioritizes: 

  • Research involving rigorous scientific methods, including for studies related to children and adolescents, where NIH is committed to approaches that reflect the highest standards of clinical care and child safety. 
  • Biological and physiological integrity: Recognizing the relevance of biological sex to health outcomes, NIH encourages applicants to account for sex-based health factors in program design, data collection, and service delivery where scientifically appropriate.
  • NIH will implement these priorities consistent with applicable laws, regulations, court orders, and all required administrative procedures. Applicants are encouraged to describe how their proposed programs align with these priorities in their project narratives. Funded activities must advance NIH's vision of protecting and improving the health and well-being of Americans. The particular focus is on those who are medically vulnerable, or live in areas with limited access to care. NIH's duty is to serve wisely, effectively, and with measurable results that justify every taxpayer dollar invested. 
Cooperative Agreement Terms and Conditions of Award

The following special terms of award are in addition to, and not in lieu of, otherwise applicable U.S. Office of Management and Budget (OMB) administrative guidelines, U.S. Department of Health and Human Services (HHS) grant administration regulations at 2 CFR Part 200, and other HHS, PHS, and NIH grant administration policies.

The administrative and funding instrument used for this program will be the cooperative agreement, an "assistance" mechanism (rather than an "acquisition" mechanism), in which substantial NIH programmatic involvement with the recipients is anticipated during the performance of the activities. Under the cooperative agreement, the NIH purpose is to support and stimulate the recipients' activities by involvement in and otherwise working jointly with the recipients in a partnership role; it is not to assume direction, prime responsibility, or a dominant role in the activities. Consistent with this concept, the dominant role and prime responsibility resides with the recipients for the project as a whole, although specific tasks and activities may be shared among the recipients and NIH as defined below.

The PD(s)/PI(s) will have the primary responsibility for:

  • Recipients will retain custody of and have primary rights to the data and software developed under these awards, subject to Government rights of access consistent with current HHS, PHS, and NIH policies.
  • Retain the primary authority and responsibility for the project as a whole and development of the methods, procedures to accomplish the aims and objectives of the NOFO, and preparation of publications.
  • Develop milestones with specific timelines and criteria for evaluation, and making appropriate revisions based on the feedback from the Principal Investigator meetings and recommendations from NIH Staff.
  • Provide, in addition to standard annual progress reports, other relevant information to NIH Staff, and coordinate and cooperate with NIH staff and other members of appropriate collaborating NIH programs.
  • Keep NIH Staff apprised of any potential impediments to execution of the goals of the Project.
  • Work directly with NIH Staff on the coordination of intra-program activities between relevant NIH programs.
  • Participate in the appropriate coordinating meetings, working groups, and/or teleconferences as needed.

NIH staff have substantial programmatic involvement that is above and beyond the normal stewardship role in awards, as described below:

  • Provide technical assistance and advice to the recipient as appropriate to achieve the aims of the cooperative agreement.
  • Help guide prioritization of datasets and knowledge base functionality development.
  • Consult on datatypes selected to be ingested, including the discussion and prioritization of datasets and datatypes to be potentially added during the duration of the award, as budget allows.
  • Monitor the progress of the Research Project, help coordinate research approaches, and contribute to the shaping of the project or approaches as warranted. NIH Staff will support and facilitate this process but will not direct it.
  • Assess and negotiate initial milestones with specific timelines and criteria for evaluation, and make appropriate revisions based on the feedback from the Principal Investigator meetings and recommendations from NIH Staff.
  • Retain the option to recommend the withholding or reduction of support from any cooperative agreement that substantially fails to achieve its goals according to the milestones agreed to at the time of the award or fails to comply with the Terms and Conditions of the award.
  • Additionally, an agency program official or IC program director will be responsible for the normal scientific and programmatic stewardship of the award and will be named in the award notice.

Areas of Joint Responsibility include:

  • NIH staff and PI(s) will convene regularly in person, virtually, or by teleconference, with additional project staff and/or NIH staff, on a schedule to be determined, to assess progress, discuss data resources, establish priorities, consider policy recommendations, and discuss strategies.
  • Collaborate to create a transition plan to aid in the transfer of data, standard operating procedures, agreements, and relationships to a third party at the conclusion of the period of performance, to continue this initiative. 
  • To enable maximum competition should this initiative be funded for another cycle, the transition plan responsibility will fall to both the PD(s)/PI(s) and NIH staff.

Dispute Resolution:

Any disagreements that may arise in scientific or programmatic matters (within the scope of the award) between recipients and NIH may be brought to Dispute Resolution. A Dispute Resolution Panel composed of three members will be convened: a designee of the recipient chosen without NIH staff voting, one NIH designee, and a third designee with expertise in the relevant area who is chosen by the other two; in the case of individual disagreement, the first member may be chosen by the individual recipient. This special dispute resolution procedure does not alter the recipient's right to appeal an adverse action that is otherwise appealable in accordance with PHS regulation 42 CFR Part 50, Subpart D and HHS regulation 45 CFR Part 16.

3. Data Management and Sharing

A Data Management and Sharing Plan (DMS Plan) is required for any NIH-funded or conducted research that will generate scientific data. Applicants must submit the DMS Plan at the time of application using the NIH DMS Plan Format Page. The DMS Plan must address the elements in the structured format. Where the DMS Plan Format Page requires a "Yes or No" response, no additional narrative is allowed. 

4. Reporting

When multiple years are involved, recipients will be required to submit the Research Performance Progress Report (RPPR) annually and financial statements as required in the NIH Grants Policy Statement Section 8.4.1 Reporting. To learn more about post-award monitoring and reporting, see the NIH Grants & Funding website, see Post-Award Monitoring and Reporting.

A final RPPR, invention statement, and the expenditure data portion of the Federal Financial Report are required for closeout of an award, as described in the NIH Grants Policy Statement Section 8.6 Closeout. NIH NOFOs outline intended research goals and objectives. Post award, NIH will review and measure performance based on the details and outcomes that are shared within the RPPR, as described at 2 CFR Part 200.301.

5. Evaluation

In carrying out its stewardship of this NOFO, the NIH or its Institutes and Centers may request information essential to an assessment of the effectiveness of this program from the participants (Center investigators, individual project investigators, and others as determined). Participants may be contacted during and after the completion of this award for periodic updates on information helpful in evaluating the impact of the program. NIH and its ICs will use this information to determine overall success of the program.
